Detection of tumor-derived extracellular vesicles in plasma from patients with solid cancer

Abstract Background Extracellular vesicles (EVs) are actively secreted by cells into body fluids and contain nucleic acids of the they originate from. The goal this study was to detect circulating tumor-derived EVs (ctEVs) mutant mRNA transcripts (EV-RNA) in plasma patients with solid cancers compare occurrence ctEVs tumor DNA (ctDNA) cell-free (cfDNA). Methods For purpose, blood from 20 15 healthy donors (HBDs) collected different preservation tubes (EDTA, BCT, CellSave) processed within 24 h venipuncture. were isolated ExoEasy protocol conditioned medium 6 cancer cell lines characterized according MISEV2018-guidelines. RNA ExoRNeasy evaluated for transcript expression levels 96 genes RT-qPCR genotyped digital PCR. Results Our workflow applied on revealed a high concordance between cellular EV-RNA as well variant allele frequencies PIK3CA , KRAS BRAF . Plasma CD9-positive EV GAPDH significantly tubes. detected only amounts (> 20%) (ctDNA). Expression profiling showed that resemble more than supporting most derived tissue.
