چکیده
Abstract Background Extracellular vesicles (EVs) are actively secreted by cells into body fluids and contain nucleic acids of the they originate from. The goal this study was to detect circulating tumor-derived EVs (ctEVs) mutant mRNA transcripts (EV-RNA) in plasma patients with solid cancers compare occurrence ctEVs tumor DNA (ctDNA) cell-free (cfDNA). Methods For purpose, blood from 20 15 healthy donors (HBDs) collected different preservation tubes (EDTA, BCT, CellSave) processed within 24 h venipuncture. were isolated ExoEasy protocol conditioned medium 6 cancer cell lines characterized according MISEV2018-guidelines. RNA ExoRNeasy evaluated for transcript expression levels 96 genes RT-qPCR genotyped digital PCR. Results Our workflow applied on revealed a high concordance between cellular EV-RNA as well variant allele frequencies PIK3CA , KRAS BRAF . Plasma CD9-positive EV GAPDH significantly tubes. detected only amounts (> 20%) (ctDNA). Expression profiling showed that resemble more than supporting most derived tissue. Conclusions We provide ctEV detection spin column-based generic isolation PCR-based measurement gene patients’ plasma. This workflow, however, tumor-specific mutations less often cfDNA.

Extracellular Vesicles Derived from Gastrointestinal Microbiota: A New Approach to Clinical Studies
Extracellular vesicles, naturally released from all cell types including bacteria, are of great importance in medical microbiology due to transporting a variety of biomaterials, enzymes, and virulence factors, regulating immunity, and having roles in colonization and initiation of signaling pathways. These vesicles are also secreted from microbiota in the gastrointestinal tract and affect the h...
متن کاملNanoplasmonic Quantification of Tumor-derived Extracellular Vesicles in Plasma Microsamples for Diagnosis and Treatment Monitoring
Tumour-derived extracellular vesicles (EVs) are of increasing interest as a resource of diagnostic biomarkers. However, most EV assays require large samples, are time-consuming, low-throughput and costly, and thus impractical for clinical use. Here, we describe a rapid, ultrasensitive and inexpensive nanoplasmon-enhanced scattering (nPES) assay that directly quantifies tumor-derived EVs from as...
متن کاملDetection of Aggregation-Competent Tau in Neuron-Derived Extracellular Vesicles
Progressive cerebral accumulation of tau aggregates is a defining feature of Alzheimer's disease (AD). A popular theory that seeks to explain the apparent spread of neurofibrillary tangle pathology proposes that aggregated tau is passed from neuron to neuron. Such a templated seeding process requires that the transferred tau contains the microtubule binding repeat domains that are necessary for...
